Chelators Packs
The in vivo imaging of biomolecules (peptides, proteins, antibodies, etc.) often requires the conjugation of a chelator prior to the radiometallation step. Recent literature has highlighted that even small modifications of the chelator can significantly impact the pharmacokinetics of the resulting conjugate. This effect is particularly pronounced for small biomolecules such as peptides or antibody fragments.
DOTA is considered a gold standard for the complexation of many radiometals. However, it is not always the optimal choice. Identifying the right combination of chelator and biovector is essential to achieve optimal in vivo performance. Key parameters such as size, geometry, lipophilicity, and charge of the metal complex can strongly influence the pharmacokinetic profile of the radiopharmaceutical.
In addition to chelator selection, the conjugation strategy itself plays a critical role in the overall performance of the bioconjugate. The choice of functional group and conjugation chemistry (e.g., NHS esters, maleimides, isothiocyanates, click chemistry) can significantly affect the stability, homogeneity, and in vivo behavior of the final construct. An inappropriate conjugation method may lead to instability or loss of targeting capability, emphasizing the importance of a well-designed bioconjugation approach.
During the early design phase of a new radiopharmaceutical, it is therefore critical to evaluate which chelator is best suited, in order to define the optimal system: chelator / radiometal / biovector.
To support this process, CheMatech offers sample packs designed to enable the rapid screening of multiple chelating agents in a cost-effective manner.
